Automated SFC Method Development: Propanolol Separation using SuperPure Discovery Analytical SFC Instrument
by Lester Dolak, PhD. and Jacquelyn Cole, Thar Technologies Inc., Pittsburgh, PA
Figure 1: SuperDiscovery AMDS System
|
The SuperPure Discovery series of analytical chromatography systems features high sample throughput and resolution to produce faster separations that meet today's demanding separation and fraction collection needs. Easy-to-use software allows maximum flexibility for control, method programming, and application-specific configurations.
Equipment and Method Setup
Figure 2: Propranolol 2 mg/mL
|
The prepared sample solution of propanolol (98%) in methanol (2 mg/mL) was placed in the autosampler. The screening method in the software was setup to automatically test the separation on various chiral stationary phases columns with different co-solvents, co-solvent percentage, and pressures. We maintained the total flow and changed the only composition of the mobile phases. The setup included equilibration between column, co-solvent, and co-solvent percentage changes.
Conclusion
Application of the automated method development capabilities of the SuperDiscovery instrument led to the separation shown. The resolution took less than 2 minutes, showed complete resolution, and sharp peaks.
